Photometric method It is a colorimetric method (colorimetric method) using photoelectric photometer. In general, the concentration of the test solution is obtained by calculating the blindness of the test solution to the transmittance of 100% or absorbance of 0, and then reading the absorbance of the test solution and calculating the ratio of the absorbance of the test solution to the concentration of the standard solution. In this case, when the relationship between the absorbance of the test solution and the concentration does not comply with the Lambart-Beer law, prepare a series of standard solutions of stepwise concentration and plot the corresponding points of the absorbance and concentration on a graph to obtain the concentration from the standard curve. This method is the most frequently used method for routine clinical chemistry.

광전광도법 광전광도계를 사용해서 하는 비색분석법(比色分布法)이다. 일반적인 실시방법은 피검액의 맹검(盲檢)을 투과율 100% 또는 흡광도 0에 맞추고 이어서 피검액의 흡광도를 읽고 똑같은 조작으로 얻은 농도기지의 표준액 흡광도와의 비례계산에 의해서 피검액의 농도를 구한다. 이 경우, 피검액의 흡광도와 농도의 관계가 람바트비어의 법칙에 따르지 않을 때에는 단계적 농도의 표준액 계열을 작성하고 그 흡광도와 농도의 대응점을 그래프로 플롯해서 생기는 표준곡선에서 농도를 구한다. 이 법은 일상임상화학검사에 가장 자주 사용되는 방법이다.
