Senescence is the aging process of living things - when cells stop dividing to replace other cells that, for some reason, stop metabolizing. Senescent cells stop dividing when telomeres, which protect the ends of chromosomes, shorten too much until they reach the Hayflick Limit - in each cell division, the telomeres lose a part and shorten, when they reach a minimum size the chromosomes stop replicating, preventing the correct division of the cell. Another interesting aspect of cell senescence is overexpression and accumulation of the beta-galactosidase enzyme in cells. These phenomena were first observed in 1995 by Dimri et al., Who proposed that a new beta-galactosidase isoform with optimal activity at pH 6.0 would exist. Even quantitative assays have been developed to detect this possible variant of the enzyme,. It is now known that what actually occurs in these cells is the accumulation of beta-gal in lysosomes and that this process is not necessary for the onset of senescence.

Senescência é o processo de envelhecimento dos seres vivos - quando as células deixam de se dividir para substituírem outras células que, por alguma razão, deixaram de metabolizar. As células senescentes deixam de dividir-se quando os telômeros, que protegem as extremidades dos cromossomos, se encurtam demais até alcançar o Limite de Hayflick - em cada divisão celular, os telômeros perdem uma parte e se encurtam, quando atingem um tamanho mínimo os cromossomos deixam de se replicar, impedindo a divisão correta da célula. Um outro aspecto interessante da senescência celular é a superexpressão e acúmulo da enzima beta-galactosidase nas células. Esse fenômenos foi inicialmente observado em 1995 por Dimri e colaboradores, os quais propuseram que existiria uma nova isoforma da beta-galactosidase com atividade ótima em pH 6.0 . Até mesmo ensaios quantitativos foram desenvolvidos para detectar essa possível variante da enzima, . Hoje sabe-se que na verdade o que ocorre nessas células é o acúmulo de beta-gal nos lisossomos e que esse processo não é necessário para o aparecimento da senescência .
