Akrosom
Das Akrosom ist eine Organelle, die sich über die vordere Hälfte des Kopfes in den Spermatozoen vieler Tiere entwickelt. Es ist eine kappenartige Struktur, die aus dem Golgi-Apparat stammt. Die Akrosomenbildung wird während der Hodenreifung abgeschlossen. Bei eutherischen Säugetieren enthält das Akrosom Verdauungsenzyme. Diese Enzyme brechen die äußere Membran der Eizelle, genannt die Zona Pellucida, so dass der haploide Kern in der Spermien Zelle mit dem haploiden Kern in der Eizelle beitreten. Dieses Ausscheiden der Akrosom- oder Akrosom-Reaktion kann in vitro durch Substanzen stimuliert werden, die eine Samenzelle auf natürliche Weise wie Progesteron oder Follikelflüssigkeit sowie das häufigere Calcium-Ionophor A23187 treffen kann. Dies kann getan werden, um als eine positive Kontrolle bei der Beurteilung der Akrosomreaktion einer Spermaprobe durch Durchflusszytometrie oder Fluoreszenzmikroskopie zu dienen. Dies geschieht in der Regel nach der Färbung mit einem fluoreszierten Lektin wie FITC-PNA, FITC-PSA, FITC-ConA oder fluoresziertem Antikörper wie FITC-CD46. Menschliches Spermatozoon Im Falle der Globozoospermie wird der Golgi-Apparat nicht in das Akrosom umgewandelt, was die männliche Unfruchtbarkeit verursacht.
