Avidin
Avidin ist ein tetrameres oder dimeres biotinbindendes Protein, das in den Eileitern von Vögeln, Reptilien und Amphibien produziert wird und in den Weißen ihrer Eier abgelagert wird. In Hühnereiweiß macht Avidin etwa 0,05% des Gesamtproteins aus. Das tetramerische Protein enthält vier identische Untereinheiten, von denen jede an Biotin mit einem hohen Grad an Affinität und Spezifität binden kann. Die Dissoziationskonstante von Avidin wird als KD ≈ 10-15 M gemessen und ist damit eine der stärksten bekannten nicht-kovalenten Bindungen. In seiner tetrameren Form wird Avidin auf eine Größe von 66-69 kDa geschätzt. 10% des Molekulargewichts werden dem Kohlenhydratgehalt aus vier bis fünf Mannose und drei N-Acetylglucosaminresten zugeschrieben. Die Kohlenhydratreste von Avidin enthalten mindestens drei einzigartige Oligosaccharid-Strukturtypen, die in ihrer Struktur und Zusammensetzung ähnlich sind. Funktionelles Avidin findet sich nur in rohem Ei, da die Biotin-Avidität des Proteins durch Kochen zerstört wird. Die natürliche Funktion von Avidin in Eiern ist nicht bekannt, obwohl es postuliert wurde, um in der Eileiter als bakterielle Wachstumsinhibitor gemacht werden, durch die Bindung von Biotin der Bakterien brauchen.
