Immunhistochemie
Immunhistochemie oder IHC bezieht sich auf den Prozess des Nachweises von Antigenen in Zellen eines Gewebeschnitts durch Ausnutzung des Prinzips von Antikörpern, die spezifisch an Antigene in biologischen Geweben binden. IHC hat seinen Namen von den Wurzeln "Immuno", in Bezug auf Antikörper, die in der Prozedur verwendet werden, und "histo", dh Gewebe. Das Verfahren wurde konzipiert und erstmals von Dr. Albert Coons im Jahre 1941 umgesetzt. Immunhistochemische Färbung ist weit verbreitet in der Diagnose von abnormen Zellen wie die in Krebs-Tumoren gefunden. Spezifische molekulare Marker sind charakteristisch für bestimmte zelluläre Ereignisse wie Proliferation oder Zelltod. IHC ist auch weit verbreitet in der Grundlagenforschung verwendet, um die Verteilung und Lokalisierung von Biomarkern und differentiell exprimierten Proteinen in verschiedenen Teilen eines biologischen Gewebes zu verstehen. Die Visualisierung einer Antikörper-Antigen-Interaktion kann auf verschiedene Weise erfolgen. Im häufigsten wird ein Antikörper an ein Enzym, wie Peroxidase, konjugiert, das eine farbgebende Reaktion katalysieren kann. Alternativ kann der Antikörper auch mit einem Fluorophor, wie Fluorescein oder Rhodamin, versehen werden.
