Microsome
In der Zellbiologie sind Mikrosomen vesikelähnliche Artefakte, die aus Stücken des endoplasmatischen Retikulums wieder gebildet werden, wenn eukaryotische Zellen im Labor aufgelöst werden; Definitionsgemäß sind Mikrosomen gewöhnlich in lebenden Zellen vorhanden. Mikrosomen können durch Differenzialzentrifugation konzentriert und von anderen Zelltrümmern getrennt werden. Ungebrochene Zellen, Kerne und Mitochondrien sedimentieren bei 10.000 g, während lösliche Enzyme und zersplitterte ER, die Cytochrom P450 enthalten, in Lösung bleiben. Bei 100.000 g, erreicht durch schnellere Zentrifugenrotation, ER-Sedimente aus der Lösung als Pellet, aber die löslichen Enzyme bleiben im Überstand. Auf diese Weise wird Cytochrom P450 in Mikrosomen konzentriert und isoliert. Mikrosomen haben eine rötlich-braune Farbe, aufgrund der Anwesenheit des eisenhaltigen Co-Faktors, Häm, in den P450s. P450s sind sehr reich an Lebern von Ratten, Mäusen und Menschen. Mikrosomen sind ein wertvolles Instrument zur Untersuchung des Stoffwechsels von Verbindungen und zur Untersuchung von Arzneimittelwirkungen durch In-vitro-Forschung. Forscher wählen oft mikrosome Lose auf der Grundlage der Enzymaktivität von spezifischen CYPs.
