Nekrose
Nekrose ist eine Form der Zellverletzung, die zu einem vorzeitigen Tod von Zellen im lebenden Gewebe durch Autolyse führt.Necrose wird durch Faktoren verursacht, die außerhalb der Zelle oder des Gewebes sind, wie Infektion, Toxine oder Trauma, die zum unregulierten Verdau von Zellkomponenten führen . Im Gegensatz dazu ist die Apoptose eine natürlich vorkommende programmierte und zielgerichtete Ursache des zellulären Todes. Während Apoptose oft nützliche Auswirkungen auf den Organismus, Nekrose ist fast immer schädlich und kann tödlich sein. Zellen, die aufgrund von Nekrose sterben, folgen nicht dem apoptotischen Signaltransduktionsweg, sondern es werden vielmehr verschiedene Rezeptoren aktiviert, die zu einem Verlust der Zellmembranintegrität und einer unkontrollierten Freisetzung von Produkten des Zelltods in den intrazellulären Raum führen. Dies initiiert in dem umliegenden Gewebe eine entzündliche Reaktion, die nahe gelegene Phagozyten daran hindert, die toten Zellen durch Phagozytose zu lokalisieren und zu eliminieren. Aus diesem Grund ist es oft notwendig, nekrotisches Gewebe chirurgisch zu entfernen, ein Verfahren, das als Debridement bekannt ist. Unbehandelte Nekrose führt zu einem Aufbau von zerlegendem totem Gewebe und Zelltrümmer an oder nahe dem Ort des Zelltods. Ein klassisches Beispiel ist Gangrän.
