Plasmid
Ein Plasmid ist ein kleines DNA-Molekül, das physisch getrennt ist und unabhängig von der chromosomalen DNA innerhalb einer Zelle replizieren kann. Am häufigsten als kleine kreisförmige, doppelsträngige DNA-Moleküle in Bakterien gefunden, sind Plasmide manchmal in archaea und eukaryotischen Organismen vorhanden. In der Natur tragen Plasmide Gene, die dem Überleben des Organismus zugute kommen können, und können häufig über einen horizontalen Gentransfer von einem Bakterium zum anderen übertragen werden. Künstliche Plasmide werden weithin als Vektoren bei der molekularen Klonierung verwendet, um die Replikation von rekombinanten DNA-Sequenzen in Wirtsorganismen zu treiben. Der Begriff Plasmid wurde erstmals 1952 vom amerikanischen Molekularbiologen Joshua Lederberg eingeführt. Plasmidgrößen variieren von 1 bis über 1.000 kbp. Die Anzahl der identischen Plasmide in einer einzigen Zelle kann unter Umständen irgendwo von einem bis zu tauschen. Plasmide können in allen drei Hauptdomänen gefunden werden: Archaea, Bakterien und Eukarya. Plasmide werden als Replikone betrachtet, die in der Lage sind, autonom innerhalb eines geeigneten Wirts zu replizieren. Jedoch werden Plasmide, wie Viren, nicht von einigen als eine Form des Lebens betrachtet.
