Reverse Transkriptase ist ein Enzym, das verwendet wird, um komplementäre DNA aus einer RNA-Matrize zu erzeugen, ein Verfahren, das als reverse Transkription bezeichnet wird. RT wird für die Replikation von Retroviren benötigt, und RT-Inhibitoren werden weithin als antiretrovirale Medikamente verwendet. RT-Aktivität ist auch mit der Replikation von Chromosomenenden und einigen mobilen genetischen Elementen verbunden. Retrovirale RT hat drei aufeinanderfolgende biochemische Aktivitäten: ▪ RNA-abhängige DNA-Polymerase, ▪ Ribonuklease H und ▪ DNA-abhängige DNA-Polymerase. Diese Aktivitäten werden von dem Retrovirus verwendet, um einzelsträngige genomische RNA in doppelsträngige cDNA umzuwandeln, die sich in das Wirtsgenom integrieren können und möglicherweise eine Langzeitinfektion erzeugen, die sehr schwer zu beseitigen ist. Die gleiche Sequenz von Reaktionen wird im Labor häufig verwendet, um RNA in DNA zur Verwendung bei der molekularen Klonierung, der RNA-Sequenzierung, der Polymerasekettenreaktion oder der Genomanalyse umzuwandeln.
