Luciferases are enzymes that catalyze biological reactions by transforming chemical energy into light energy. Like all enzymes, luciferase has high specificity to its substrate, increases the speed and decreases the activation energy of the reaction, has high catalytic activity and is not consumed in the process. The Enzymes Commission of the International Union of Biochemistry classified luciferase in the group of Oxirredutases, which are enzymes that catalyze oxidation reactions between two substrates. The catalysis performed by luciferase acts in the bioluminescence process. Luciferase occurs predominantly in marine organisms; in the terrestrial environment it is found mainly in the class of insects, which is the richest in bioluminescent species. Luciferase is found in about 2000 species in the orders Diptera, Collembola and mainly Coleoptera. The first purification of insect luciferase occurred in the 1950s, being the North American lamina Photinus pyralis. To about 20 years the cDNA for the luciferase of this firefly. was cloned, sequenced and the primary structure of the protein was deduced. A Comissão de Enzimas da União Internacional de Bioquímica classificou a luciferase no grupo das Oxirredutases, que são enzimas que catalisam as reações de oxirredução entre dois substratos. A catalisação realizada pela luciferase atua no processo de bioluminescência. A luciferase ocorre predominantemente em organismos marinhos, no ambiente terrestre é encontrada principalmente na classe dos insetos, que é a mais rica em espécies bioluminescentes. A luciferase é encontrada em cerca de 2000 espécies nas ordens Diptera, Collembola e principalmente Coleoptera. A primeira purificação de luciferase de insetos ocorreu na década de 50, sendo do lampirídeo norte-americano Photinus pyralis. A cerca de 20 anos o cDNA para a luciferase deste vagalume. foi clonado, sequenciado e a estrutura primaria da proteína foi deduzida.
