Ultramicroscope
Un ultramicroscope est un microscope avec un système d'éclairage qui permet de visualiser de minuscules particules. Lorsque le diamètre d'une particule est inférieur ou proche de la longueur d'onde de la lumière visible, la particule ne peut pas être vue dans un microscope optique avec la méthode d'illumination habituelle. Le système ultramicroscope est basé sur la diffusion de la lumière, et non sur la réflexion de la lumière. Dans le système, les particules à observer sont dispersées dans un colloïde liquide ou gazeux. Le colloïde est placé dans une enceinte sombre absorbant la lumière et éclairé par un faisceau convergent de lumière intense entrant d'un côté. La lumière qui frappe les particules colloïdales sera dispersée. Dans les discussions sur la diffusion de la lumière, le faisceau convergent s'appelle un «cône Tyndall». La scène est vue par un microscope ordinaire placé perpendiculairement à la direction du faisceau lumineux. Au microscope, les particules individuelles apparaîtront comme de petites taches floues de lumière qui se déplacent irrégulièrement. Les taches sont intrinsèquement floues car la diffusion de la lumière produit des images plus floues que la réflexion de la lumière. Les particules sont dans le mouvement brownien dans la plupart des types de colloïdes liquides et gazeux, ce qui provoque le mouvement des taches.
