Il fattore intrinseco, noto anche come fattore intrinseco gastrico, è una glicoproteina prodotta dalle cellule parietali dello stomaco. È necessario per l'assorbimento di vitamina B12 in seguito nell'intestino tenue. Nell'uomo, la proteina del fattore intrinseco gastrico è codificata dal gene GIF. All'ingresso nello stomaco, la vitamina B12 si lega all'ottacrocina, una glicoproteina. Il complesso risultante entra nel duodeno, dove enzimi pancreatici digeriscono l'acidotracina. Nell'ambiente meno acido del piccolo intestino, B12 può quindi legarsi al fattore intrinseco. Questo nuovo complesso si muove all'atrio, dove cellule epiteliali speciali li endocitano. All'interno della cella, B12 si dissociata ancora una volta e si lega ad un'altra proteina, il transcobalamina II. Il nuovo complesso può uscire dalle cellule epiteliali per entrare nel fegato.
