Anemia megaloblastica
L'anemia megaloblastica (o anemia megaloblastica) è un'anemia (di classificazione macrocitica) che risulta dall'inibizione della sintesi del DNA durante la produzione di globuli rossi. Quando la sintesi del DNA è compromessa, il ciclo cellulare non può progredire dalla fase di crescita di G2 alla fase della mitosi (M). Ciò porta alla continua crescita cellulare senza divisione, che presenta come macrocitosi. L'anemia megaloblastica ha un inizio piuttosto lento, soprattutto se paragonata a quella di altre anemie. Il difetto della sintesi del DNA a cellule rosse è spesso dovuto all'ipovolaminosi, in particolare una carenza di vitamina B12 e / o acido folico. La carenza di vitamina B12 da solo non provoca la sindrome in presenza di folato sufficiente, poiché il meccanismo è la perdita del riciclaggio folico dipendente da B12, seguito da una perdita di folato della sintesi dell'acido nucleico (in particolare la timoina), causando difetti nella sintesi del DNA. L'integrazione dell'acido folico in assenza di vitamina B12 impedisce questo tipo di anemia (anche se altre patologie specifiche della vitamina B12 continuano). La perdita di micronutrienti può anche essere una causa.
