Nestorius fu arcivescovo di Costantinopoli dal 10 aprile 428 fino all'agosto 431, quando l'imperatore Teodosio II confermò la sua condanna dal Consiglio di Efeso il 22 giugno. I suoi insegnamenti includevano un rifiuto del lungo uso del titolo di Theotokos, "Madre di Dio", per Maria, madre di Gesù, e furono fraintesi da molti affinché non credesse che Cristo fosse veramente Dio. Questo lo ha portato in conflitto con altri importanti uomini del tempo, in particolare Cyril di Alessandria, che accusava di eresia. Nestorio cercò di difendersi al primo Concilio di Efeso nel 431, ma invece si trovò formalmente condannato per eresia da una maggioranza dei vescovi e successivamente rimosso dalla sua vista. Su sua richiesta si ritirò nel suo antico monastero in o vicino ad Antiochia. Nel 435 il Teodosio II lo mandò in esilio nell'Alto Egitto, dove visse fino al 450, difendendo con forza la sua ortodossia. Il suo ultimo difensore principale nell'Impero Romano, Theodoret di Cirro, finalmente acconsentì a metterlo in atto nel 451 durante il Concilio di Calcedonia; Da allora non aveva difensori nell'impero. Ma la Chiesa dell'Est non ha mai accettato la sua condanna.
