Un plasmide è una piccola molecola del DNA che è fisicamente separata da e può replicare indipendentemente dal DNA cromosomico all'interno di una cellula. Più comunemente si trovano come piccole molecole DNA circolari, a doppio filamento, nei batteri, talvolta sono presenti plasmidi in archea e organismi eucariotici. In natura, i plasmidi trasportano geni che possono beneficiare della sopravvivenza dell'organismo e spesso possono essere trasmessi da un batterio all'altro attraverso il trasferimento genico orizzontale. I plasmidi artificiali sono ampiamente usati come vettori nella clonazione molecolare, in grado di guidare la replicazione delle sequenze di DNA ricombinanti all'interno degli organismi ospiti. Il termine plasmide è stato introdotto per la prima volta dal biologo molecolare americano Joshua Lederberg nel 1952. Le dimensioni dei plasmidi variano da 1 a più di 1.000 kbp. Il numero di plasmidi identici in una sola cellula può variare da una a migliaia in alcune circostanze. I plasmidi possono essere trovati in tutti e tre i principali domini: Archaea, Bacteria e Eukarya. I plasmidi sono considerati repliconi, capaci di replicare autonomamente all'interno di un host adeguato. Tuttavia, i plasmidi, come i virus, non sono considerati da alcuni come una forma di vita.
