Trascrittasi inversa
La transcriptasi inversa è un enzima usato per generare DNA complementare da un modello di RNA, un processo chiamato trascrizione inversa. RT è necessaria per la replicazione dei retrovirus e gli inibitori della RT sono ampiamente usati come farmaci antiretrovirali. L'attività RT è associata anche alla replica delle estremità cromosomiche e di alcuni elementi genetici mobili. RT retrovirale ha tre attività biochimiche sequenziali: ▪ DNA polimerasi dipendente dall'RNA, ▪ ribonucleasi H, e ▪ polimerasi DNA-dipendente. Queste attività sono usate dal retrovirus per convertire l'RNA genomico a singolo filamento in cDNA a doppio filamento che può integrarsi nel genoma ospite, generando potenzialmente un'infezione a lungo termine che può essere molto difficile da sradicare. La stessa sequenza di reazioni è ampiamente usata in laboratorio per convertire l'RNA in DNA per l'impiego in clonazione molecolare, sequenziamento di RNA, reazione a catena della polimerasi o analisi genomica.
