Nella chimica, la spettrofotometria è la misura quantitativa delle proprietà di riflessione o trasmissione di un materiale in funzione della lunghezza d'onda. È più specifico della spettroscopia elettromagnetica generale in quella spettrofotometria che tratta di luce visibile, quasi ultravioletta e vicino a infrarossi, ma non copre tecniche spettroscopiche risolte in tempo. La spettrofotometria comporta l'utilizzo di uno spettrofotometro. Uno spettrofotometro è un fotometro che può misurare l'intensità in funzione della lunghezza d'onda della sorgente luminosa. Le caratteristiche importanti degli spettrofotometri sono la larghezza di banda spettrale e l'intervallo lineare di misurazione dell'assorbimento o della riflessione. Uno spettrofotometro è comunemente usato per misurare la trasmittanza o la riflessione di soluzioni, solidi trasparenti o opachi, come vetro lucidato o gas. Tuttavia, essi possono anche essere progettati per misurare la diffusività su uno qualsiasi dei campi luminosi elencati che di solito coprono circa 200 nm - 2500 nm utilizzando diversi controlli e calibrazioni. All'interno di questi campi di luce sono necessarie tarature sulla macchina usando standard che variano a seconda della lunghezza d'onda della determinazione fotometrica.
