타목시펜은 활성 대사 산물 인 4- 하이드 록시 타목시펜을 통해 유방 조직에서 에스트로겐 수용체의 길항제입니다. 자궁 내막과 같은 다른 조직에서는 작용제로 작용하므로 혼합 된 작용제 / 길항제로 특징 지을 수 있습니다. 타목시펜은 폐경 전 여성에서 호르몬 수용체 양성 유방암에 대한 일반적인 내분비 치료제이며, 폐경기 이후 여성에서도 표준으로 사용됩니다. 아로마 타제 억제제가 빈번히 사용됩니다. 일부 유방암 세포는 에스트로겐이 필요합니다. 에스트로겐은 이들 세포에서 에스트로겐 수용체에 결합하고이를 활성화시킵니다. 타목시펜은 에스트로겐 수용체에도 결합하지만 활성화시키지 않는 화합물로 대사됩니다. 이 경쟁적인 길항 작용 때문에 타목시펜은 다른 키가 삽입되는 것을 방지하는 자물쇠에서 깨진 열쇠처럼 작동하여 에스트로겐이 수용체에 결합하는 것을 방지합니다. 따라서 유방암 세포의 성장이 차단됩니다. Tamoxifen is an antagonist of the estrogen receptor in breast tissue via its active metabolite, 4-hydroxytamoxifen. In other tissues such as the endometrium, it behaves as an agonist, and thus may be characterized as a mixed agonist/antagonist. Tamoxifen is the usual endocrine therapy for hormone receptor-positive breast cancer in pre-menopausal women, and is also a standard in post-menopausal women although aromatase inhibitors are also frequently used in that setting. Some breast cancer cells require estrogen to grow. Estrogen binds to and activates the estrogen receptor in these cells. Tamoxifen is metabolized into compounds that also bind to the estrogen receptor but do not activate it. Because of this competitive antagonism, tamoxifen acts like a key broken off in the lock that prevents any other key from being inserted, preventing estrogen from binding to its receptor. Hence breast cancer cell growth is blocked. Tamoxifen was discovered by pharmaceutical company Imperial Chemical Industries and is sold under the trade names Nolvadex, Istubal, and Valodex.
