Kromosom polytene adalah kromosom yang berukuran lebih besar yang telah berkembang dari kromosom piawai dan biasanya terdapat dalam kelenjar salivary melroskopi Drosophila. Sel-sel khusus menjalani replikasi DNA berulang tanpa pembahagian sel, untuk meningkatkan jumlah sel, membentuk kromosom polytene gergasi. Kromosom polytene terbentuk apabila pelbagai pusingan replikasi menghasilkan banyak kromatid kakak yang masih tersambung bersama.
