Anemia megaloblástica
A anemia megaloblástica (ou anemia megaloblástica) é uma anemia (de classificação macrocítica) que resulta da inibição da síntese do DNA durante a produção de glóbulos vermelhos. Quando a síntese de DNA é prejudicada, o ciclo celular não pode avançar do estágio de crescimento de G2 para o estágio de mitosis (M). Isso leva ao contínuo crescimento celular sem divisão, que se apresenta como macrocitose. A anemia megaloblástica tem um início bastante lento, especialmente quando comparado com o de outras anemias. O defeito na síntese de DNA de células vermelhas é mais frequentemente devido a hipovitaminose, especificamente uma deficiência de vitamina B12 e / ou ácido fólico. A deficiência de vitamina B12 por si só não causará a síndrome na presença de folato suficiente, pois o mecanismo é a perda de reciclagem de folato dependente de B12, seguido de perda de deficiência de folato de síntese de ácido nucleico (especificamente timina), levando a defeitos na síntese de DNA. A suplementação de ácido fólico na ausência de vitamina B12 previne esse tipo de anemia (embora outras patologias específicas de vitamina B12 continuem). A perda de micronutrientes também pode ser uma causa.
