Metachromasy
A metacromásia é uma mudança característica na cor da coloração realizada em tecidos biológicos, exibida por certos corantes de anilina quando se ligam a substâncias particulares presentes nesses tecidos, chamados cromotrópicos. Por exemplo, o azul de toluidina fica cor-de-rosa quando ligado à cartilagem. A ausência de mudança de cor na coloração é chamada de orto-cromossas. O mecanismo subjacente à metachromasia é a presença de polianões dentro do tecido. Quando estes tecidos são corados com uma solução de corante básica concentrada, tal como azul de toluidina, as moléculas de corantes estão próximas o suficiente para formar agregados diméricos e poliméricos. As propriedades de absorção destas agregações diferem das das moléculas de corantes individuais não agregadas. As estruturas de células e tecidos que possuem altas concentrações de grupos sulfato e fosfato ionizados - tais como a substância moída da cartilagem, grânulos que contém heparina de mastócitos e reticulo endoplasmático áspero das células plasmáticas - apresentam metachromasia. Portanto, o azul de toluidina aparecerá roxo a vermelho quando ele manchar esses componentes.
