Primeiro, isolado em 1964, em sua forma pura, pelos cientistas israelenses Raphael Mechoulam e Yechiel Gaoni no Weizmann Institute of Science, é um sólido vítreo transparente à água quando frio, que se torna viscoso e pegajoso se aquecido. Uma formulação farmacêutica de (-) - trans-Δ9-tetrahidrocannabinol, conhecida por seu dronabinol INN, está disponível por prescrição nos EUA e no Canadá sob a marca Marinol. Um terpenoide aromático, THC tem uma solubilidade muito baixa em água, mas boa solubilidade na maioria dos solventes orgânicos, especificamente lipídios e álcoois. Como a maioria dos metabolitos secundários farmacologicamente ativos das plantas, o THC na cannabis é suposto estar envolvido na autodefesa, talvez contra os herbívoros. THC também possui altas propriedades de absorção de UV-B (280-315 nm), que, especularam, poderiam proteger a planta da exposição prejudicial à radiação UV.
