В биохимии лигаза представляет собой фермент, который может катализировать соединение двух крупных молекул путем образования новой химической связи, как правило, при сопутствующем гидролизе небольшой химической группы, зависящей от одной из больших молекул или фермента, катализирующего соединение двух соединений, например, ферменты, которые катализируют соединение CO, CS, CN и т. д. В общем случае лигаза катализирует следующую реакцию: Ab + C → A-C + b или иногда Ab + cD → A-D + b + c, где нижний регистр буквы обозначают маленькие зависимые группы. Лигаза может соединяться с двумя комплементарными фрагментами нуклеиновой кислоты и восстанавливать одноцепочечные разрывы, которые возникают в ДНК с двойной цепью во время репликации.
