Kooperatiflik, bireysel unsurların bağımsız olarak hareket ettikleri hipotetik bir standart etkileşimsiz sisteme göre birbirinden bağımsız olarak hareket eden özdeş ya da hemen hemen aynı elemanları içeren sistemler tarafından görüntülenen bir olgudur. Bunun bir tezahürü, bağlayıcı bölgelerin bir ligand için afinitesinin arttığı, pozitif kooperatifliğe veya azaltılmış, negatif kooperatifliğe sahip bir enzimin veya reseptörlerin, bir ligandın bir bağlama sitesine bağlanmasından sonra oluştuğu noktalardır. Örneğin, oksijen için hemoglobin'in dört bağlanma bölgesinin afinitesi, ilk oksijen molekülü bağlandığında bağlanmamış hemoglobininkinden daha fazla artmaktadır. Buna kooperatif bağlama denir. Aynı zamanda pek çok özdeş altbirimden oluşan büyük zincir moleküllerde, bu moleküllerin erime, açılma veya çözülme gibi faz geçişlerine uğradıklarında kooperatifliği görüyoruz. Buna alt birim kooperatifliği denir. Bununla birlikte, "enerji" kavramının daima standart bir devletle ilişkili olarak tanımlanması gerektiği için, ardışık ligand bağlama adımlarına afinitede belirgin bir artış veya azalmaya dayanan kooperatifçilik tanımlaması sorunludur.
