Fumaraz, fümaratın malat haline geri dönüşümlü hidrasyon / dehidrasyonunu katalize eden bir enzimdir. Fumaraz iki şekilde gelir: mitokondrial ve sitosolik. Mitokondriyal izoenzim Krebs Döngüsünde bulunur ve sitozolik izoenzim, amino asitler ve fumarat metabolizmasına katılır. Subhücresel lokalizasyon, mitokondriyal formdaki amino terminus üzerinde bir sinyal sekansı varlığında kurulurken, sitosolik formdaki subselüler lokalizasyon, mitokondriyal çeşitte bulunan sinyal dizisinin yokluğu ile kurulur. Bu enzim 3 metabolik yola katılır: sitrik asit döngüsü, indirgeyici sitrik asit döngüsü ve renal hücreli karsinom. Bu gendeki mutasyonlar, böbrek hücreli karsinom ile birlikte deri ve rahimde leiomyomaların gelişimi ile ilişkilendirilmiştir.
