Leptin, "tokluk hormonu", vücutta depolanan yağ miktarını regüle eden yağ hücreleri tarafından yapılan bir hormondur. Bunu hem açlığın hissiyatını hem de enerji harcamalarını ayarlayarak yapar. Açlık, depolanan yağ miktarı belirli bir seviyeye ulaştığında engellenir. Leptin daha sonra salgılanır ve vücuda dolaşır, sonunda hipotalamusun kavisli çekirdeğinde leptin reseptörlerini aktive eder. Enerji harcamaları hem beyindeki sinyalle hem de direkt olarak çevresel hedefler üzerindeki leptin reseptörleri ile artar. Leptinin etkisi "açlık hormonu" olan grelinin tersidir. Ghrelin reseptörleri, leptin reseptörleri ile aynı beyin hücrelerinde bulunur, bu yüzden hücreler rakip tokluk ve açlık sinyalleri alırlar. Leptin ve grelin, birçok başka hormon ile birlikte, enerji homeostazının karmaşık sürecine katılırlar.
