Faz kontrast mikroskopisi
Faz kontrast mikroskopisi, şeffaf bir örneğin içinden geçen ışıktaki faz kaymalarını, görüntüdeki parlaklık değişimlerine çeviren optik bir mikroskopi tekniğidir. Faz kaydırmaları kendilerini görünmez, ancak parlaklık varyasyonları olarak gösterildiğinde görülebilir hale gelir. Şekil 1: Aynı hücreler geleneksel parlak alan mikroskobu ile ve faz kontrast mikroskopisi ile görüntülendi. Işık dalgaları vakum dışındaki bir ortamdan geçtiğinde, ortamla etkileşim, dalga amplitüd ve fazının ortamın özelliklerine bağlı olarak değişmesine neden olur. Genlikteki değişiklikler çoğunlukla dalga boyuna bağlı olan ve renklere yol açabilen ışığın saçılmasından ve absorbe edilmesinden kaynaklanır. Fotoğraf ekipmanı ve insan gözü yalnızca genlik varyasyonlarına duyarlıdır. Özel düzenlemeler olmadan faz değişiklikleri bu nedenle görünmezdir. Bununla birlikte, faz değişiklikleri çoğu zaman önemli bilgiler taşır. Faz kontrast mikroskopisi biyolojide özellikle önemlidir. Şekil 1'de örneklendiği gibi basit bir parlak alan mikroskopu ile görünmeyen birçok hücresel yapı ortaya koymaktadır.
