Hemoglobin (kısaltma: Hb), oksijen ve karbondioksit ile birleşerek kan dolaşımı sistemi yoluyla vücut dokularına taşınan kırmızı kan hücrelerindeki bir protein molekülüdür. Hemoglobin içindeki Fe + 2 formundaki demir iyonu kanda kırmızı kan verir. Normal koşullar altında, 100 ml kan, 0.03 gram oksijen taşıyabilen 15 gram hemoglobin içerir. Kandaki hemoglobin içeriğini ölçmenin çeşitli yolları vardır, çoğunlukla kana karşı bazı testler yapmak için özel olarak tasarlanmış makineler tarafından otomatik olarak yapılır. Bu makine içerisinde, eriyik halindeki hemoglobin izole etmek için kırmızı kan hücreleri ayrılır. Bu serbest hemoglobin, cyanmethemoglobin oluşturmak için hemoglobin moleküllerine sahip güçlü siyanür içeren kimyasallarla karıştırılır. Işık bir cyanmethemoglobin çözeltisi ile aydınlatılarak emilen ışık miktarını ölçerek (özellikle 540 nanometre dalga için), hemoglobin sayısı belirlenebilir. Hemoglobin seviyesi genellikle her dekaliter için gram cinsinden hemoglobin sayısı (gm) olarak belirlenir (100 mililitre).
