Histon

Histones are basic proteins that occur in the nucleus of eukaryotes. They are essential as a component of the chromatin for the packaging of the DNA, but also for the expression of some of the encoded genes. In order to accommodate the large genome of the eukaryotes in the cell nucleus, it is necessary to package the DNA particularly. In the case of eukaryotes, this occurs in chromosomes whose smallest packing unit is a nucleosome. A nucleosome is an "octamer" consisting of eight histones: two copies each of the histones H2A, H2B, H3 and H4. The DNA is wound 1.65 times by a nucleosome, which corresponds to a DNA length of 146 base pairs. The non-associated DNA that binds adjacent nucleosomes is called linker DNA. The histone H1 binds DNA directly next to nucleosomes and allows the next higher packing unit of the DNA. Histones consist of a globular center and flexible terminal arms, which have many basic, ie positively charged amino acids. The DNA, on the other hand, is negatively charged, so that there is electrostatic attraction. Histone sind basische Proteine, die im Zellkern von Eukaryoten vorkommen. Sie sind als Bestandteil des Chromatins für die Verpackung der DNA, aber auch für die Expression mancher auf ihr codierten Gene von essentieller Bedeutung. Um das große Genom der Eukaryoten im Zellkern unterbringen zu können, ist es nötig, die DNA besonders zu verpacken. Bei Eukaryoten geschieht dies in Chromosomen, deren kleinste Verpackungseinheit ein Nukleosom ist. Ein Nukleosom ist ein „Oktamer“, es besteht aus acht Histonen: je zwei Kopien der Histone H2A, H2B, H3 und H4. Die DNA ist 1,65-mal um ein Nukleosom gewickelt, was einer DNA-Länge von 146 Basenpaaren entspricht. Die nicht assoziierte DNA, die benachbarte Nukleosomen verbindet, wird Linker-DNA genannt. Das Histon H1 bindet DNA direkt neben Nukleosomen und erlaubt die nächsthöhere Verpackungseinheit der DNA. Histone bestehen aus einem globulären Zentrum und flexiblen endständigen Armen, die viele basische, also positiv geladene Aminosäuren besitzen. Die DNA ist hingegen negativ geladen, so dass eine elektrostatische Anziehung besteht.
