Currently, this is rarely quoted. 측쇄설 엘리히 Ehrlich에 의하여 생각하여 낸 항체생성(抗體)의 기전이다. 이 설은, 항독소(抗毒素)의 연구를 기반으로 하는 것으로, 생체세포의 표면에 독소와 결합하는 반응부위, 즉 측쇄(側鎖)가 존재하고, 독소는 이것을 통하여 세포에 독작용을 미친다고 생각하였다. 그 위에 독소의 작용에 견디어 살아남은 세포는 이의 측쇄(側鎖)가 과잉으로 생산되어 여분의 측쇄(側鎖)는 혈중에 유리(遊離)되어 이것이 항독소항체로서 독소와 결합하여 독작용을 중화한다고 생각하였다. 현재 이설은 거의 인용되고 있지 않다.
