Nigrosin
La nigrosina es una mezcla de colorantes negros sintéticos fabricados calentando una mezcla de nitrobenceno, anilina y clorhidrato de anilina en presencia de un catalizador de cobre o hierro. Sus principales usos industriales son como colorantes para lacas y barnices y tintas para rotuladores. La sulfonación de nigrosina produce un colorante aniónico soluble en agua, nigrosina WS. En biología, la nigrosina se usa para la tinción negativa de bacterias, así como el hongo que contiene cápsulas, Cryptococcus neoformans Las formas y tamaños de los organismos se ven como contornos libres de color contra el fondo oscuro. Una ventaja de usar este método, en lugar de manchas positivas regulares como el azul de metileno o la fucsina de carbol, es que la fijación previa por calor o alcohol no es necesaria, por lo que los organismos se ven en formas más realistas. Además, la tinción negativa con nigrosina puede revelar algunos microorganismos que no pueden ser manchados por métodos regulares. Nigrosin WS se utiliza en las pruebas de viabilidad: las células vivas excluyen el colorante, pero entra en las células muertas.
