Canal ionique à voie ligandaire
Les canaux ioniques à voie ligand sont un groupe de protéines de canaux ioniques transmembranaires qui s'ouvrent pour permettre aux ions tels que Na +, K +, Ca2 + ou Cl- de passer à travers la membrane en réponse à la liaison d'un messager chimique tel qu'un neurotransmetteur. Ces protéines sont généralement composées d'au moins deux domaines différents: un domaine transmembranaire qui comprend le pore ionique et un domaine extracellulaire qui comprend l'emplacement de liaison du ligand. Cette modularité a permis une approche «diviser et conquérir» pour trouver la structure des protéines. La fonction de ces récepteurs situés dans les synapses est de convertir directement le signal chimique du neurotransmetteur pré-amorti directement et très rapidement en un signal électrique postsynaptique. De nombreux LGIC sont en outre modulés par des ligands allostériques, par des inhibiteurs de canaux, des ions ou le potentiel de la membrane. Les LGIC sont classés en trois superfamilies qui n'ont pas de relation évolutive: les récepteurs Cys-loop, les récepteurs Ionotropic glutamate et les chaînes ATP-gated. Les LGIC peuvent être contrastés avec les récepteurs métabotropes, les canaux ioniques à tension et les canaux ioniques activés par étirage.
