Nella genetica, una croce di prova, introdotta per la prima volta da Gregor Mendel, comporta l'allevamento di un individuo caratteristico dominante con un individuo recessivo, al fine di determinare la zigosità del primo analizzando le proporzioni di prole con il fenotipo recessivo. Il genotipo che una prole ha per ciascuno dei suoi geni è determinata dall'allele ereditato dai suoi genitori. La combinazione di alleli è il risultato dei cromosomi materni e paterni che hanno contribuito da ogni gamete alla fecondazione di quella discendenza. Durante la meiosi nei gameti, i cromosomi omologhi sperimentano la ricombinazione genetica e si separano casualmente nelle cellule figlia aposloide, ognuna con una combinazione unica di geni codificati maternamente e paternamente. Le croci di prova implicano l'allevamento dell'individuo in questione con un altro individuo che esprime una versione recessiva dello stesso carattere. Se tutte le prole mostrano il fenotipo dominante, l'individuo in questione è omozigote dominante; Se la prole presenta sia fenotipi dominanti che recessivi, allora l'individuo è eterozigote.
