プロドラッグの投与後に生体内で目的とする化合物に変化されること定義される。 有用な薬物にもかかわらず、副作用、安定性、溶解性、吸水性、作用時間など適さない性質を持っていること、化学的修飾を加えて、臨床使用可能にしたものである。 例えば、胃腸障害、多くの非ステロイド抗炎症薬であるプロドラッグが開発されているが、不活性物質として、消化管からの吸収され、その後、肝臓などで代謝されて活性型の物質に変化して抗炎症作用を発揮することで胃腸障害の軽減が 期待されている。 프로드러그 투여후에 생체내에서 목적으로 하는 화합물로 변화되는 것으로 정의된다. 유용한 약물임에도 불구하고 부작용, 안정성, 용해성, 흡수성, 작용시간등에서 적합치 않는 성질을 가지고 있는 것에 화학적 수식을 가해서 임상사용 가능하게 한 것이다. 예를 들면 위장장애가 많은 비스테로이드 항염증약인 프로드러그가 개발되고 있는데, 불활성 물질로서 소화관에서 흡수되고 그 후 간장등에서 대사되어서 활성형의 물질로 변화해서 항염증 작용을 발휘하는 것으로 위장장애의 경감이 기대되고 있다.
