Na biologia molecular, o termo dupla hélice refere-se à estrutura formada por moléculas de cadeia dupla de ácidos nucleicos, como o DNA. A estrutura de dupla hélice de um complexo de ácido nucleico surge como conseqüência de sua estrutura secundária e é um componente fundamental na determinação da sua estrutura terciária. O termo entrou na cultura popular com a publicação em 1968 de The Double Helix: uma conta pessoal da descoberta da estrutura do DNA, de James Watson. O polímero de dupla hélice de DNA de ácidos nucleicos, mantido em conjunto por nucleotídeos que unem pares de bases. No B-DNA, a estrutura dupla helicoidal mais comum, a dupla hélice é diestra com cerca de 10-10,5 nucleótidos por turno. A estrutura de dupla hélice do DNA contém uma ranhura principal e uma ranhura menor, sendo a ranhura principal maior do que a ranhura menor. Dada a diferença nas larguras do sulco principal e do sulco menor, muitas proteínas que se ligam ao DNA fazem isso através do sulco principal mais largo.
