Methylphenidat ist ein substituiertes Phenethylamin und ein psychostimulierendes Arzneimittel, das zur Behandlung von Aufmerksamkeitsdefizit-Hyperaktivitätsstörung, posturielles orthostatisches Tachykardie-Syndrom und Narkolepsie verwendet wird. Methylphenidat wurde seit über 50 Jahren untersucht und recherchiert und hat eine sehr gute Wirksamkeit und Sicherheit für die Behandlung von ADHS. Das ursprüngliche Patent war im Besitz von CIBA, jetzt Novartis Corporation. Es wurde zum ersten Mal von der US-amerikanischen Food and Drug Administration im Jahr 1955 für die Behandlung, was damals bekannt als Hyperaktivität lizenziert. Vorgeschrieben für Patienten ab 1960, wurde die Droge stark in den 1990er Jahren verschrieben, als die Diagnose von ADHS selbst wurde weithin akzeptiert. ADHD und andere ähnliche Bedingungen sind vermutlich mit der Unterleistung der Dopamin- und Norepinephrin-Funktionen im Gehirn verbunden, vor allem in der präfrontalen Kortex, die für die Selbstregulierungsfunktion und die Exekutivfunktion verantwortlich sind. Der Wirkmechanismus von Methylphenidat beinhaltet die Hemmung der Catecholamin-Wiederaufnahme, vor allem als Dopamin-Wiederaufnahmeinhibitor.
