Kjeldahl법은 종래부터 표준법으로 되어 있으나 조작이 복잡하고, 단백의 종류에 따라 질소량에서 단백량으로의 환산계수(換算係數)에 상당한 차가 있다. 뷰렛법은 조작이 간단하고, 그 발색(發色)은 단백의 종류에 따른 차가 적으며, 일상의 임상검사에 적합한 방법이다. 또 자동화법으로 여러 형(型)의 장치에 적용되고 있다. 굴절률법은 정확도(正確度)는 약간 떨어지지만, 실시가 간편하여 널리 응용되고 있다. 정상치는 6.7~8.3g/dl로서, 탈수(脫水), 세망내피계질환, 만성전염병일 때 증가하고, 혈장단백질의 누출, 영양불량, 간기능장애일 때 감소한다.
