En genética, una prueba cruzada, introducida por primera vez por Gregor Mendel, implica la cría de un rasgo dominante individual con un individuo recesivo, con el fin de determinar la cigosidad de la primera analizando proporciones de descendencia con el fenotipo recesivo. El genotipo que una descendencia tiene para cada uno de sus genes está determinado por el alelo heredado de sus padres. La combinación de alelos es el resultado de los cromosomas maternos y paternos aportados por cada gameta en la fertilización de esa descendencia. Durante la meiosis en gametos, los cromosomas homólogos experimentan recombinación genética y se segregan aleatoriamente en células hija haploides, cada una con una combinación única de genes codificados maternal y paternalmente. Los cruces de prueba implican criar al individuo en cuestión con otro individuo que expresa una versión recesiva del mismo rasgo. Si todos los descendientes muestran el fenotipo dominante, el individuo en cuestión es homocigótico dominante; Si los descendientes muestran fenotipos tanto dominantes como recesivos, entonces el individuo es heterocigota.
