Azathioprine
L'azathioprine est un médicament immunosuppresseur utilisé dans la transplantation d'organes et les maladies auto-immunes et appartient à la classe chimique des analogues de la purine. Synthétisé à l'origine comme un médicament contre le cancer et un promédicament pour la mercaptopurine en 1957, il a été largement utilisé comme immunosuppresseur depuis plus de 50 ans. L'azathioprine agit comme un promédicament pour la mercaptopurine, inhibant une enzyme qui est nécessaire pour la synthèse de l'ADN. Ainsi, il affecte le plus fortement les cellules proliférantes, telles que les cellules T et les cellules B du système immunitaire. Le principal effet néfaste de l'azathioprine est la suppression de la moelle osseuse, ce qui peut mettre la vie en danger, en particulier chez les personnes présentant une déficience génétique de l'enzyme thiopurine S-méthyltransférase. Il est également répertorié par l'Agence internationale de recherche sur le cancer en tant que cancérogène du groupe 1.
