În genetică, o mutație missense este o mutație punctuală în care o singură schimbare de nucleotide are ca rezultat un codon care codifică un aminoacid diferit. Mutațiile mușchilor pot transforma proteina rezultată nefuncțională, iar astfel de mutații sunt responsabile pentru bolile cum ar fi buloasa epidermoliză, boala celulelor seceră și ALS mediată de SOD1. Un tip special de mutație de tip missense care duce la trunchierea codului este mutația nonsens în care un codon este schimbat la un codon de stop. Cea mai comună variantă a bolii celulă seceră, cea de-a 20-a nucleotidă a genei pentru lanțul beta al hemoglobinei, este modificată de la codonul GAG la GUG. Astfel, acidul glutamic al celui de-al 6-lea aminoacid este înlocuit cu valina - notată ca o mutație "E6V" - și proteina este suficient de alterată pentru a provoca boala celulei secera. Nu toate mutațiile deranjante duc la apariția unor modificări semnificative ale proteinelor. Un aminoacid poate fi înlocuit cu un aminoacid cu proprietăți chimice foarte asemănătoare, caz în care proteina poate funcționa normal; aceasta se numește o mutație neutră, "silențioasă", "silențioasă" sau conservatoare.

In genetics, a missense mutation is a point mutation in which a single nucleotide change results in a codon that codes for a different amino acid. Missense mutations can render the resulting protein nonfunctional, and such mutations are responsible for diseases such as Epidermolysis bullosa, sickle-cell disease, and SOD1 mediated ALS. A special type of missense mutation that results in truncation of code is the nonsense mutation in which a codon is changed to a stop codon. The most common variant of sickle-cell disease, the 20th nucleotide of the gene for the beta chain of hemoglobin is altered from the codon GAG to GUG. Thus, the 6th amino acid glutamic acid is substituted for valine — notated as an "E6V" mutation — and the protein is sufficiently altered to cause the sickle-cell disease. Not all missense mutations lead to appreciable protein changes. An amino acid may be replaced by an amino acid of very similar chemical properties, in which case, the protein may still function normally; this is termed a neutral, "quiet", "silent" or conservative mutation.
