Prophase, din anticul grecesc pre și fazis, este o etapă de mitoză în care cromatina se condensează în structuri în formă de tijă dublă numite cromozomi în care cromatina devine vizibilă. Acest proces, numit condensarea cromatinei, este implicat în complexul de condensat. Deoarece materialul genetic a fost reprodus în interfața anterioară a ciclului celular, există două copii identice ale fiecărui cromozom în celulă. Aceste copii se numesc cromatide sora și sunt atașate una de alta la un element ADN prezent pe fiecare cromozom numit centromere. De asemenea, în timpul profazei, poate fi aplicată colorarea giemsa pentru a provoca bandă G în cromozomi. O organelle importantă în mitoză este centrozomul, centrul de organizare a microtubulelor în metazoani. În timpul profazei, cele două centrozomi, care se repetă independent de mitoză, au crescut activitatea microtubulelor datorită recrutării de tubulină γ. Centrosomii vor fi împinși spre capetele opuse ale nucleului celular prin acțiunea motoarelor moleculare care acționează asupra microtubulelor. Practic, cromatina din nucleul se învârte în cromozomi.
