Auxotrophie

The auxotrophs are organisms which can not synthesize certain essential substances independently. Thus, they are dependent on absorbing these substances from the environment. An example of this are lactic acid bacteria, which, because of their specialization, are dependent on nutrient-rich media and are no longer able to synthesize many substances that are necessary for themselves. If an organism is auxotrophic for a substance, this mutant, for example in the case of adenine auxotrophy, is referred to as "Ade-". The opposite of auxotrophy is called prototrophy. These are organisms which are not dependent on suppline, that is, they can synthesize all necessary substances themselves. Auxotrophies often occur as mutations in the genome due to mutagenesis. If an auxotrophic mutant is transformed with a vector carrying the missing functional gene, the intact gene can compensate for the auxotrophic defect. The mutant is then again able to synthesize the substance itself. In this case one speaks of a complementation. Als auxotroph bezeichnet man Organismen, die bestimmte essentielle Substanzen nicht selbstständig synthetisieren können. Somit sind sie darauf angewiesen, diese Stoffe aus der Umwelt aufzunehmen. Ein Beispiel dafür sind Milchsäurebakterien, die aufgrund ihrer Spezialisierung auf nährstoffreiche Medien angewiesen sind und viele lebensnotwendige Stoffe nicht mehr selbst synthetisieren können. Ist ein Organismus auxotroph für eine Substanz wird diese Mutante, zum Beispiel im Falle einer Adenin Auxotrophie, als "Ade−" bezeichnet. Das Gegenteil von Auxotrophie wird Prototrophie genannt. Es handelt sich dabei um Organismen, die nicht auf Suppline angewiesen sind, also alle benötigten Stoffe selbst synthetisieren können. Auxotrophien treten häufig infolge von Mutagenese als Mutationen im Genom auf. Transformiert man eine auxotrophe Mutante mit einem Vektor, der das fehlende funktionsfähige Gen trägt, so kann das intakte Gen den auxotrophen Defekt ausgleichen. Die Mutante ist dann wieder in der Lage, den Stoff selbst zu synthetisieren. In diesem Fall spricht man von einer Komplementation.
