Lorsqu'il est oxydé, il forme de l'hémateine, un composé qui forme des complexes fortement colorés avec certains ions métalliques, les plus notoires étant des sels de Fe (III) et Al (III). Les complexes métal-hématéine sont utilisés pour tacher les noyaux cellulaires avant l'examen au microscope. Les structures qui tachent avec l'hématine de fer ou d'aluminium sont souvent appelées basophiles, même si le mécanisme de la coloration est différent de celui de la coloration avec des colorants basiques. L'hématoxyline et l'éosine forment ensemble l'hématoxyline et la tache d'éosine, l'une des taches les plus utilisées dans l'histologie. C'est une tache permanente par opposition aux taches temporaires (par exemple, solution d'iode dans KI). Une autre tache commune est l'hématoxyline d'acide phosphotungstique, un mélange d'hématoxyline avec de l'acide phosphotungstique. Au début des années 1970 et en 2008, il y avait pénurie d'hématoxyline en raison de l'interruption de l'extraction de bois de feuillus. When oxidized it forms haematein, a compound that forms strongly coloured complexes with certain metal ions, the most notable ones being Fe(III) and Al(III) salts. Metal-haematein complexes are used to stain cell nuclei prior to examination under a microscope. Structures that stain with iron- or aluminium-haematein are often called basophilic, even though the mechanism of the staining is different from that of staining with basic dyes. Haematoxylin and eosin together make up haematoxylin and eosin stain, one of the most commonly used stains in histology. It is a permanent stain as opposed to temporary stains (e.g. iodine solution in KI). Another common stain is phosphotungstic acid haematoxylin, a mix of haematoxylin with phosphotungstic acid. In the early 1970s and in 2008, there were shortages of haematoxylin due to interruptions in its extraction from logwood. The price of the compound increased, affecting the cost of diagnostic histopathology, and prompted a search for alternative nuclear stains.
