În medicină, o comă este o stare de inconștiență care durează mai mult de șase ore, în care o persoană: nu poate fi trezită; nu răspunde în mod normal la stimuli, lumină sau sunet dureroase; nu are un ciclu normal de somn-trezire; și nu inițiază acțiuni voluntare. O persoană aflată în stare de comă este descrisă ca fiind comă. O persoană comatată prezintă o lipsă totală de veghe și nu este capabilă să simtă, să vorbească, să audă sau să se miște în mod conștient. Pentru ca un pacient să-și mențină conștiința, trebuie să funcționeze două componente neurologice importante. Primul este cortexul cerebral - materia cenușie care acoperă stratul exterior al creierului. Celălalt este o structură situată în brainstem, numită sistem de activare reticulară. Leziuni la una sau ambele componente sunt suficiente pentru a determina un pacient să experimenteze o comă. Cortexul cerebral este un grup de materie cenușie densă, densă, compusă din nucleul neuronilor a căror axoni formează apoi "materia albă" și este responsabil de percepție, releu al intrării senzoriale prin calea talamică și mulți alte funcții neurologice, inclusiv gândirea complexă.
