Malignité est la tendance d'un état de santé à devenir progressivement pire. La malignité est la plus connue comme caractérisation du cancer. Une tumeur maligne contraste avec une tumeur bénigne non cancéreuse dans la mesure où une malignité n'est pas auto-limitée dans sa croissance, est capable d'envahir des tissus adjacents et peut être capable de se propager à des tissus éloignés. Une tumeur bénigne n'a aucune de ces propriétés. La cancérologie est caractérisée par une anaplasie, une invasivité et une métastase. Les tumeurs malignes sont également caractérisées par une instabilité du génome, de sorte que les cancers, évalués par séquençage du génome complet, ont souvent entre 10 000 et 100 000 mutations dans l'ensemble de leur génome. Les cancers montrent généralement une hétérogénéité tumorale, contenant plusieurs sous-clones. Ils ont également souvent une expression réduite des enzymes de réparation de l'ADN en raison de la méthylation épigénétique des gènes de réparation de l'ADN ou des microARN modifiés qui contrôlent l'expression du gène de réparation de l'ADN.
